Does anyone know of forskolin (Coleus Forskolii extract) being used to treat Parkinson's disease?


Classic Parkinson's Disease (PD) involves a shortage of dopamine due to the death of dopamine-producing neurons in the Substantia Nigra (SN) portion of the brain.

Forskolin (FSK) is a small, fat-soluble molecule and so may be able to cross the blood-brain barrier.

FSK activates adenylate cyclase (AC).

Activated AC produces more cyclic AMP (cAMP).

cAMP activates Protein Kinase A (PKA).

Activated PKA phosphorylates Tyrosine Hydroxylase (TH).

TH is the substance in SN neurons that, when active, produces L-DOPA, which is converted into dopamine.

TH becomes inactive when catecholamines (dopamine, epinephrine or norepinephrine) attach to it.

When phosphorylated, TH can shake off attached catecholamines and become active again, resulting in more L-DOPA, and hence more dopamine.

I am theorizing that taking FSK as a supplement could result in more L-DOPA, and hence could relieve or erase symptoms of PD. However, I have not been able to find any example of FSK being used to treat PD.
