How do pain relievers work?


I know they do something with blood vessels and nerve endings, but if you have a headache, why if you took a pain reliever designed for temporary relief, will the pain never come back?

If the pain doesn't come back either A) the pill had a placebo effect on you and your body is tricked into thinking it feels better and therefore does or B) the pain was relatively minor and by resting the muscles(or nerves or whatever hurt) it fought off the cause of the stress
